Design Notes
To avoid intron complications (as H. jecorina is a fungi), the amino acid sequence was copied from NCBI (protein P62694.1) and imported into the DNA 2.0 software. The sequence was then back-translated under E. coli optimization. There were Pst1 (ctgcag) sites at bp 67, which was mutated to ctgcaa, and at bp 1239, which was mutated to ctgctc. There were EcoR1 sites (gaattc) at bp 406, which was mutated to gaattt, and at bp 427, which was mutated to gaattt. As there were no stop codons on this modified sequence, two stop codons were added: taataa.
